It is somewhat ironic that animals that are the prime choice for detailed genetic analysis, such as the fruit fly and the nematode, have thus far been largely refractory to reverse genetic analysis. Their detailed genetic map, and small genome size have made them subjects of ambitious genome analysis projects, but there is still no strategy to introduce desired changes into their genomes by homologous recombination. Some alternative approaches have recently become available; this review describes possibilities and unsolved problems for (...) reverse genetics in the nematode Caenorhabditis elegans. Many of the ten practices to abolish war of just peacemaking theory can be appropriated by classical realist thinkers to illumine possibilities of more peace for the post-cold war situation. The optimism of just peacemaking theory about abolishing war, however, does not need to be appropriated. Realist participation in the just peacemaking project can proceed but only with reservations about what seems to be a mixture of optimism and Kantian idealism about the future peacefulness of a capitalist world, and the (...) illusion that war will disappear from the world. Realism, grounded more in the prophets than the just peacemaking project and more in the prophets' moral critique than in Thucydides' cynicism, provides a stronger foundation for policy advice than the Sermon on the Mount which did not focus on international relations. The striking lack of attention by Jesus to questions of the management of the Roman Empire and the ethics of war and peace permits Christians to consult books of the Bible where international relations and foreign policy are prominent for moral wisdom on the subject. U'IWversity of Scranton Scranton, Pennsylvania EDWARD VACEK shrewdly observes that proportionalism attempts to synthesize the crucial insights of both the teleologist and the deontologist.1 Indeed, Vacek provides a fine summary of this achievement. However, he reflects that the most underdeveloped feature of proportionalism is its value theory by which we are enabled to know how (...) to resolve conflict situations.2 Richard McCormick, among others, has attempted to meet this deficiency by ·adopting John Finnis' notion of associated basic goods. In this essay, I will show how such an ·approach fails to remain true to the basic aims of proportionalism. Nor should this come as a surprise, since Finnis is himself indebted to one of McCormick's archopponents, Germain Grisez. Moreover, I will attempt to outline the elements involved in a non-hierarchfoal theory of value which I will argue to be the only adequate alternative to McCormick 's attempted solution. Deontologists argue that certain acts are right or wrong regardless of the ·consequences. Proportionalists, however, are opposed to such notions as an " intrinsically evil " act or an "absolute" duty, since they recognize the existence of moral dilemmas. A moral dilemma arises whenever there is a situation in which pre-moral values •are in conflcit, i.e., whatever choice one makes will result in the omission or harm of one or other of the values at stake. McCormick's initial description of the requirements of proportionate reason [s ias follows: "a) a v1alue at least equal to that sacrificed is at 1 stake; b) there is no less harmful way of protecting ithe value here and now; c) the manner of its protection here and now will not undermine it in the long run." 8 McCormick argues that it simply will not do to claim that the ontic evil permitted or caused in a conflict situation is always only indirectly intended.4 For, in many instances, this simply is not true. Moreover, he would argue that to intend an end is not necess1arily to desire it as deontologists suppose. What justifies the ontic evil resulting from such actions, then, is solely the proportionate reason for which it is allowed or caused. There are some obvious criticisms that have been hurled at this proportionalist proposal for resolving conflict situations: I) that it leaves itself open to the practical, if not theoretical, difficulty of weighing different values;!2) that it results in the possibility of many resolutions that could go counter to our normal moral intuitions; 8) that there exists an inherent fallibility in the calcul,ation of long-range consequences; and 4) that this uncertainty might easliy promote an undesirable attitude of skeptical relativism. This first book-length study of Paul Tillich's ethics is drawn from research in the Harvard Archives and fifty years of teaching Tillich's social-political thought. In Ronald H. Stone's fourth work on Tillich's philosophy the ethic is examined from the early ontological to socialist ethics to his own final principled-situationalist ethic in late life. Unique to this study is the in-depth inquiry into Tillich's courageous social action correlated with his own philosophical-theological ethic. The Political Crisis and Christian Ethics addresses themes in political philosophy in the context of a crisis in democracy after the denial of the 2020 election by the Republican candidate for president. The refusal to accept the results of the election divided the electorate and drove the president's followers to fail in their attempted coup attempt in January of 2020. Democracy is defended in Reinhold Niebuhr's writing on politics and in Barack Obama's use of the theologian's thought. It is developed (...) further in the political theory of Paul Tillich. In spite of Stein and Glasier's justifiable conclusion that initial optimism concerning the immediate clinical applicability of neural transplantation was premature, there exists much experimental evidence to support the potential for incorporating this procedure into a therapeutic arsenal in the future. To realize this potential will require continued evolution of our knowledge at multiple levels of the clinical and basic neurosciences.

It may seem to their opponents that they are trying to have their cake and eat it too. Postmodernists admit that their own paradigm must be and will be placed into question by future thinkers. But if they can anticipate an eventual reaffirmation of their paradoxical stand in an ongoing oscillating debate, then cannot it be said that they have arrived at a truth that transcends their time and place in history? And, if so, is not their fallibilist stance in (...) fact self-referentially inconsistent? The response of postmodernists is the claim that each reaffirmation of a fallibilist epistemology and ethics throughout history is in fact sui generis. And this is the case because each reaffirmation has its own unique context within which it is made. Modernists will of course argue that these contextual differences are nonessential and irrelevant. And the debate over the problem of the One and the Many is once again launched in a new context. Thus far from running away from the paradoxical position that what they assert is both true and false, postmodernists revel in such inconsistencies.But does not such an ethical stance resemble the Sisyphean nightmare of being condemned to roll a heavy stone up a cliff only to have it keep falling back to the bottom ad infinitum? If no decision is innocent of doing some harm in the world, why should we bother to play the moral game at all? Indeed, what possible help is a postmodernist ethics when it comes to making some of the complex and crucial decisions we face today if it refuses to say anything substantial beyond the recommendations that we be cautious and balanced?And the postmodernist can only reply that we are letting our neurotic need for solid foundations frighten us. For ethics is an art not a science. There are no absolute rules. If we do not like the way the game is set up, then we are simply revealing our ultimate hubris in the face of a mystery requiring deep humility. (shrink)

This comprehensive educational history of public schools in Hawai'i shows and analyzes how dominant cultural and educational policy have affected the education experiences of Native Hawaiians. Drawing on institutional theory as a scholarly lens, the authors focus on four historical cases representing over 150 years of contact with the West. They carefully link historical events, significant people, educational policy, and law to cultural and social consequences for Native Hawaiian children and youth. The authors argue that since the early 1800s, educational (...) policy in Hawai'i emphasizing efficiency has resulted in institutional structures that have degenerated Hawaiian culture, self-image, and sovereignty. Native Hawaiians have often been denied equal access to quality schools and resulting increased economic and social status. These policies were often overtly, or covertly, racist and reflected wider cultural views prevalent across the United States regarding the assimilation of groups into the American mainstream culture. The case of education in Hawai'i is used to initiate a broader discussion of similar historical trends in assimilating children of different backgrounds into the American system of education. The scholarly analysis presented in this book draws out historical, political, cultural, and organizational implications that can be employed to understand other Native and non-Native contexts. Given the increasing cultural diversity of the United States and the perceived failure of the American educational system in light of these changes, this book provides an exceptionally appropriate starting point to begin a discussion about past, present, and future schooling for our nation's children. Because it is written and comes from a Native perspective, the value of the "insider" view is illuminated. This underlying reminder of the Native eye is woven throughout the book in _Ha'awina No'ono'o_--the sharing of thoughts from the Native Hawaiian author. With its primary focus on the education of native groups, this book is an extraordinary and useful work for scholars, thoughtful practitioners, policymakers, and those interested in Hawai'i, Hawaiian education, and educational policy and theory. Electron micrographs of active ribosomal genes from many species show a similar picture in which gene regions covered with nascent transcripts alternate with apparently non‐transcribed spacers. Since the gradients of visible nascent transcripts stop near the 3′ end of the 28S sequence it has often been assumed that transcription by RNA polymerase I also terminates at that point. Recent biochemical studies have shown however, that transcription continues far beyond the 3′ end of the 28S and in some species continues across (...) the entire spacer. In this article we review the evidence for spacer transcription in Xenopus, mouse and Drosophila and discuss possible reasons for its invisibility in the electron microscope. (shrink)

In the summer of 1997 one could scarcely enter a bookstore in Beijing without encountering Wang Xiaobo's pensive and defiant look on the cover of dozens of books displayed at the entrance. Wang had suddenly died in the spring of that year at the age of forty-five. Born in Beijing in 1952 to a family of intellectuals, he remained attached to China's capital despite periods of separation, such as during the Cultural Revolution, when he was sent to Yunnan to "learn (...) from the peasants" and taught in a "people-run-school" in Shandong, and also during the 1980s, when he studied in the United States . Wang always returned to Beijing, in the late 1970s to study economy and business at the People's University and in the late 1980s to teach there. After retiring in 1993, he devoted his time to writing: poetry, novels, essays, non-fiction, and a movie script. (shrink)

The challenge of understanding our existence and our place in the universe has perplexed humans for centuries. What we must remember, according to Ronald Dufty, is that the environment of the earth is a very small region of the universe, and that it is continually evolving. From this basis, Dufty assesses commonly held scientific and philosophical beliefs, and seeks to correct the misconceptions they lead to. Dufty considers topical issues such as global warming, religious terrorism and anti-social behaviour with (...) a constant awareness of the bigger picture and a desire to expand our knowledge of the nature of the universe and our role in it. Despite a long period of neglect, research on emotion in organizational behavior has developed into a major field over the past 15 years, and is now seen to be part of an affective revolution in the organization sciences. In this article, we review current research on emotion in the organizational behavior field based on five levels of analysis: within person, between persons, dyadic interactions, leadership and teams, and organization-wide. Specific topics we cover include affective events theory, state and trait affect (...) and mood, emotional intelligence, emotional labor, emotional contagion, emotions and leadership, and building a healthy emotional climate. Rakowski's form of egalitarianism places him squarely within the dominant individualistic liberal tradition developed over the past two or three decades by John Rawls, Ronald Dworkin, and others. He calls his view "equality of fortune," and states its "central thesis" this way: "no one should have less valuable resources and opportunities available to him than anyone else, simply in virtue of some chance occurrence the risk of which he did not choose to incur". Those who suffer from the fallout (...) of the natural lottery have "a right to compensation" from "fortune's favorites". Thus, his answer to the question Amartya Sen has popularized --"Equality of what?"--is not, contrary to appearances, equality of fortune. Smith’s constant concern with the problems of secularization made the subject of the memorial volume most appropriate. The wide respect for Professor Smith’s thought, his visiting appointments in both Europe and the United States, and the fact that Scotland has long served as a theological bridge between European and Anglo-Saxon interests accounts for essays by S. A dynamic view of history as the avenue for speaking of God in a secular age is the common theme. Although the essays concentrate on theological concerns with secularization, Nicol and Templeton make extensive use of Collingwood’s philosophy of history as an approach to problems of history and transcendence. Controversy persists over the ethics of compensating research participants and providing posttrial benefits to communities in developing countries. Little is known about residents' views on these subjects. In this study, interviews about compensation and posttrial benefits from a hypothetical HIV vaccine trial were conducted in Uganda’s Rakai District. Most respondents said researchers owed the community posttrial benefits and research compensation, but opinions differed as to what these should be. Debates about posttrial benefits and compensation rarely include residents' views like these, (...) but future ones should. (shrink)

We describe here recent work on the molecular genetics of mitosis in the filamentous fungus Aspergillus nidulans. Aspergillus is one of three simple eukaryotes with powerful genetic systems that have been used to analyze mitosis. The modern molecular biological techniques available with this organism have made it possible to use mutations to identify genes and proteins that play an important role in mitosis. Three Aspergillus genes that affect mitosis are described. One gene, nimA, is specifically expressed late in the cell (...) cycle and codes for a putative protein kinase that induces mitosis, even in cells blocked in S‐phase. The second gene, bimG, codes for a putative phosphatase that interacts functionally with the nimA kinase. The third gene, bimE, codes for a protein that suppresses mitosis during interphase, apparently by keeping nimA turned off. None of these genes appear to be similar to any of the genes affecting mitosis that have been characterized in other eukaryotes, but rather appear to be elements of a system that prevents mitosis from occurring during interphase. The discovery of induced pluripotent stem (iPS) cells in 2006 was heralded as a major breakthrough in stem cell research. Since then, progress in iPS cell technology has paved the way towards clinical application, particularly cell replacement therapy, which has refueled debate on the ethics of stem cell research. However, much of the discourse has focused on questions of moral status and potentiality, overlooking the ethical issues which are introduced by the clinical testing of iPS cell replacement therapy. First-in-human trials, (...) in particular, raise a number of ethical concerns including informed consent, subject recruitment and harm minimisation as well as the inherent uncertainty and risks which are involved in testing medical procedures on humans for the first time. These issues, while a feature of any human research, become more complex in the case of iPS cell therapy, given the seriousness of the potential risks, the unreliability of available animal models, the vulnerability of the target patient group, and the high stakes of such an intensely public area of science. Our paper will present a detailed case study of iPS cell replacement therapy for Parkinson's disease to highlight these broader ethical and epistemological concerns. If we accept that iPS cell technology is fraught with challenges which go far beyond merely refuting the potentiality of the stem cell line, we conclude that iPS cell research should not replace, but proceed alongside embryonic and adult somatic stem cell research to promote cross-fertilisation of knowledge and better clinical outcomes. In the management and business ethics literatures, stakeholder engagement has been demonstrated to lead to more ethical management practices. However, there may be limits on the extent to which stakeholder engagement can, as currently conceptualized, resolve some of the more difficult ethical challenges faced by managers. In this paper we argue that stakeholder engagement, when seen as a way of reducing five types of knowledge problems—risk, ambiguity, complexity, equivocality, and a priori irreducible uncertainty—can aid managers in resolving such ethical challenges. Using a practical illustration of the ethical challenges surrounding the development and application of genetic modification technologies, we demonstrate how stakeholder engagement enables managers to better address these knowledge problems, thereby to manage more ethically. Oligonucleotide‐directed, site‐specific mutagenesis is being applied to the problem of ion‐gradient driven active transport with the lac permease as a model system. It has been shown that Arg‐302, His‐322 and Glu‐325, neighboring residues in putative transmembrane helices IX and X, play an important role in lactose‐coupled H+ translocation, possibly as components of a catalytic triad similar to that found in the serine proteases. In addition, Cys residues, long thought to be involved in the mechanism of the permease, are not directly (...) involved in either substrate binding or H+ translocation. Finally, a variety of mutations have no effect on permease activity indicating that single amino acid changes do not lead indiscriminately to long‐range conformational alterations. (shrink)

Somatic cell gene therapy has yielded promising results. If germ cell gene therapy can be developed, the promise is even greater: hundreds of genetic diseases might be virtually eliminated. But some claim the procedure is morally unacceptable. We thoroughly and sympathetically examine several possible reasons for this claim but find them inadequate. There is no moral reason, then, not to develop and employ germ-line gene therapy. Taking the offensive, we argue next that medicine has a prima facie moral obligation to (...) do so. (shrink)
